Often it will seal by itself but I've had a 521 window replaced and the installer squirted a rubberized sealer around the outside between glass and gasket. Also between body and gasket. This stuff doesn't harden exactly and can be scrapped off and the gasket re-used if the glass gets replaced again. I've used black urethane but it hardens like cured rubber and while it seals very well it bonds the glass to the gasket.

It should be removable. The stuff the glass shop used gets firm is flexible and you can scrape it off.... the next time the glass needs changing. Most RTV silicone becomes permanent and then you would need a new gasket..

I'm saying you don't want it permanently sealed. The stuff I'm mentioning can be stripped away. It was like 'liquid rubber' that thickens to seal but can be scrapped off again. Maybe you mean the same thing.
